  <dc:title xml:lang="srp">Imunohistohemijske karakteristike i redoks status hipofizno-gonadalnog sistema nakon aplikacije anaboličkog steroida peripubertalnim i adultnim mužjacima pacova</dc:title>

Uvod: Anabolički androgeni steroidi (AAS) predstavljaju sintetske derivate muškog polnog hormona testosterona. Tokom poslednjih nekoliko decenija, zloubotreba AAS postala je široko rasprostranjena među profesionalnim ali i među rekreativnim
sportistima. Poslednjih godina, nandrolon dekanoat predstavlja jedan od najčešće
zloupotrebljavanih AAS u sportu.
Cilj: Cilj ovog istraživanja bio je da se ispitaju efekti hronične primene nandrolon
dekanoata i treninga plivanjem na histomorfometrijske parametre gonadotropnih ćelija
hipofize, histološke promene na testistu, skeletnom mišiću i masnom tkivu, kao i na
redoks status kod peripubertalnih i adultnih mužjaka pacova.
Materijal i metode: U studiju su uključene 64 životinje muškog pola (po 32 životinje
za svaku starosnu grupu) starosti 5 nedelja (peripubertalni) i starosti 10 nedelja
(adultni) svrstanih u 4 ispitivane grupe: kontrolna grupa - (T-N-), grupa koja prima
steroide (T-N+), grupa koja trenira plivanje (T+N-) i grupa koja trenira plivanje i
prima steroide (T+N+). Životinje koje su primale steroide dobijale su jednom nedeljno
nandrolon dekanoat u dozi od 20 mg/kg telesne mase, subkutano. Životinje koje su
trenirale plivanje, svakog dana su plivale 1sat /5 dana u nedelji. Nakon završenog
četvoronedeljnog eksperimentalnog procesa, životinje su žrtvovane. Izolovana tkiva
hipofize, testisa, skeletnog mišića i masnog tkiva rutinski su obrađivana i
pripremljena prema standardnim histološkim protokolima. U prikupljenim uzorcima
krvi, određivane su vrednosti testosterona, LH, FSH, leptina, insulina kao i prooskidativni markeri i enzimi sistema antioksidativne zaštite.
Rezultati: Naši rezultati su pokazali da tretman nandrolon dekanoatom značajno
smanjuje volumensku gustinu gonadotropnih ćelija hipofize u obe ispitivane starosne
populacije. Broj ćelija LH ćelija hipofize na mm2 značajno je smanjen nakon samostalne
i udružene primene nandrolona i treninga plivanjem u adultnoj populaciji mužjaka.
Serumske vrednosti LH i FSH hormona snižene su nakon tretmana nandrolonom, dok su
istovremeno vrednosti testosterona u krvi povišene. Ekspresija androgenog receptora u
testisu smanjena je nakon tretmana nandrolonom. U obe ispitivane starosne populacije
primećeno je najveće povećanje dijametra i površine skeletnog mišića nakon udružene
primene nandrolon dekanoata i treninga plivanjem. U T+N+ grupi uočeno je najveće
smanjenje dijametra i površine preseka adipocita kao i serumskih vrednosti leptina.
Povećanje vrednosti prooksidativnih markera i u tkivu i u krvi uočeno je nakon
primene nandrolona.
Zaključak: Nandrolon dekanoat samostalno i u kombinaciji sa treningom plivanjem
snižava histomorfometrijske parametre gonadotropnih ćelija hipofize, ekspresiju
androgenog receptora u testisu, serumske vrednosti LH, FSH hormona i leptina, dok
istovremeno povećava koncentraciju testosterona i dijametar skeletnih mišićnih
ćelija.</dc:description>
  <dc:description xml:lang="eng">Abstract:
Introduction: Anabolic androgenic steroids (AAS) are synthetic derivatives of the male sex hormone testosterone. During the last few decades, the abuse of AAS has become widespread among professional as well as recreational  portsman. In recent years, nandrolone decanoate has been one of the most frequently abused AASs in sports.
Aim: The aim of this study was to investigate the effects of the chronic administration of nandrolone decanoate and swimming training on histomorphometric parameters of the pituitary gonadotrophic cells, histological changes in the testis, skeletal muscle and fat tissue as well as
redox status in peripubertal and adult male rats. 
Material and methods: The study included 64 male rats (32 animals per age group) 5 weeks old (peripubertal) and 10 weeks old (adults) divided into 4 experimental groups: control group - (TN-), the group that received steroids (T-N+), swimming training group (T+N-) and swimming training with steroid administration group (T+N+). The steroid receiving animals, weekly
received nandrolone decanoate in a dose of 20 mg/kg b.w, subcutaneously. Rats that trained by swimming, swam every day 1 hour, 5 days a week. After completing the four-week experimental protocol, rats were sacrificed. Isolated pituitary gland, testicles, skeletal muscle and fat tissue
were routinely processed and prepared according to standard histological protocols. In the collected blood samples, the values of testosterone, LH, FSH, leptin, insulin, pro-oxidative markers and enzymes of the antioxidant defense system were determined.
Results: Our results have shown that the treatment with nandrolone decanoate significantly reduces the volume density of pituitary gonadotropic cells in both populations. Number of LH pituitary cells per mm2 was significantly decreased after nandrolone treatment alone or combined with swimming training in adult rats. Serum levels of LH and FSH hormone were decreased after
treatment with nandrolone while levels of testosterone were increased. Expresion of androgenic receptor in testes were decreased after treatment with nandrolone. In both population the largest increase of cross-section area and diameter of skeletal muscle cells was oberved after combined 
administration of nandrolone and swimming training. The largesed decrease of the diameter and cross-section areas of the adipocytes and serum level of leptin was observed in T+N+ group. The increased blood and tissue levels of pro-oxidative markers was observed after treatment with nandrolone.
Conclusion: Nandrolone decanoate alone or combined with swimming training decreases histomorphometric parameters of pituitary gonadotrophic cells, expresion of androgenic receptors in testes, serum levels of LH, FSH hormons and leptin. Increse of the blood testosteron levels and size of skeletal muscle cells can also be observed.</dc:description>
