  <dc:title xml:lang="srp">Ispitivanje antimikrobnih, antiinflamacijskih, antioksidacionih i kardioprotektivnih efekata ekstrakta sremuša, Allium ursinum L</dc:title>

  <dc:description xml:lang="srp">Cilj ove studije bio je ispitivanje antimikrobnih, antioksidacionih, 
antiinflamacijskih i kardioprotektivnih efekata ekstrakata lista sremuša (Allium 
ursinum L). Za određivanje antimikrobnih efekata korišćen je mikrodilucioni test, 
dok je za ispitivanje antiinflamacijskog potencijala korišćen model inflamacije 
izazvane karageninom na šapi pacova. Za ispitivanje antiinflamacijskog efekta 
uključeno je 50 pacova podeljenih u pet grupa: pacovi na tretmanu 0,9% NaCl, pacovi 
na tretmanu indometacinom i pacovi tretirani ekstraktom sremuša u tri doze-50, 
100 i 200 mg/kg intraperitonealno (i.p.). Za ispitivanje kardioprotektivnih efekata 
ekstrakta sremuša korišćen je model retrogradne perfuzije po Langendorff-u. U ovaj 
deo eksperimenta uključeno je 80 pacova podeljenih u 8 grupa: zdravi i spontano 
hipertenzivni pacovi (SHR) koji su konzumirali ekstrakt sremuša u dozi od 125 
mg/kg, 250 mg/kg i 500 mg/kg per os tokom 28 dana. U uzorcima koronarnog venskog 
efluenta određivani su nivoi prooksidanasa, dok je iz uzoraka venske krvi određivan 
sistemski redok status. Rezultati su pokazali generalno slabo antibakterijsko i 
antigljivično delovanje ekstrakata sremuša koje je bilo zavisno od vrste
mikroorganizma i tipa ekstrakta. Metanolni ekstrakt sremuša u dozi od 200 mg/kg
ispoljio je najintenzivniji efekat u redukciji inflamacije šape izazvane 
karageninom. Dodatno, unos ekstrakta sremuša tokom 28 dana značajno je doprineo 
oporavku srčane funkcije i ublažio oksidaciona oštećenja srca. Ekstrakt sremuša 
ispoljio je potencijal ka modulaciji sistemske redoks ravnoteže i istakao se kao 
snažan antioksidans. Benefiti ove biljne vrste uočeni su kako na modelu zdravih 
tako i kod SHR pacova pri čemu su izraženiji efekti bili prisutni nakon unosa 
više doze ekstrakta. Rezultati ove doktorske disertacije ukazuju na značajnu ulogu 
ekstrakta sremuša u kardioprotekciji i snažno antioksidaciono delovanje što 
ukazuje na mogućnost njegovog korišćenja samostalno u prevenciji ili kao adjuvantna 
antioksidaciona terapija kod kardiovaskularnih bolesti.</dc:description>
  <dc:description xml:lang="eng">he aim of this study was to investigate the antimicrobial, antioxidant, antiinflammatory and cardioprotective effects of Allium ursinum L extracts. A microdilution test was used to determine the antimicrobial effects, while a model of carrageenan-induced inflammation in rat paws was used to examine the anti-inflammatory potential. To examine 
the anti-inflammatory effect, 50 rats were divided into five groups: rats treated with 0.9% 
NaCl, rats treated with indomethacin and rats treated with wild garlic extract in three doses -
50, 100 and 200 mg / kg intraperitoneally (i.p.). A Langendorff retrograde perfusion model 
was used to examine the cardioprotective effects of wild garlic extract. This part of the 
experiment included 80 rats divided into 8 groups: healthy and spontaneously hypertensive 
rats (SHR) who consumed sremus extracts at a dose of 125 mg/kg, 250 mg/kg and 500 mg/kg 
per os for 28 days. Levels of pro-oxidants were determined in coronary venous effluent 
samples, while systemic redox status was determined in venous blood samples. Our results 
showed a generally weak antibacterial and antifungal effect of wild garlic extract, which was 
dependent on the type of microorganism and extract. Methanol extract of wild garlic at a dose 
of 200 mg/kg showed the most intensive effect in reducing paw inflammation caused by 
carrageenan. In addition, intake of wild garlic extract for 28 days significantly contributed to 
the recovery of cardiac function and alleviated oxidative damage to the heart. Wild garlic
extract showed the potential to modulate the systemic redox balance and stood out as a 
powerful antioxidant. Benefits of this plant species were observed in both the model of 
healthy and SHR rats, with more pronounced effects observed after intake of a higher dose of 
the extract. The results of this doctoral thesis indicate a significant role of wild garlic extract 
in cardioprotection and strong antioxidant activity, which indicates the possibility of its use 
alone in prevention or as adjuvant antioxidant therapy in cardiovascular diseases.</dc:description>
